Abstract

Bluetongue is caused by an RNA-virus belonging to the Orbivirus genus. It affects sheep, cattle and other ruminants and is transmitted by midges from the Culicoides genus. It causes important economical losses. There is a notorious difference between the 24 different BTV serotypes. The objective of this study is to analyze the distribution of the Bluetongue virus in different tissues by Reverse Transcription-Polymerase Chain Reaction (RT-PCR) for BTV serotype 1 in naturally infected sheep from Andalucía, where serotypes 1 and 4 are present. RNA was extracted from different tissues sample of these sheep and after that, a generic and serotype 1-especific RT-PCR was carried out. Only the blood sample from one of these sheep was positive and the sequence of the fragment proved that it belonged to BTV serotype 1. This demostrates that blood is a good sample for the detection of serotype 1. Further assays will be developed in order to determinate the efficacy of this RT-PCR.
